Thesis title: GENETIC VARIATION OF CERTAIN GENES RELATED TO GROWTH TRAITS IN SOME GOAT BREEDS RAISED IN EGYPT
Eight functional candidate genes for growth traits (GH1, GH2, GH6, IGF-I, IGFBP3, MSTN, PRL and BMP-15) polymorphisms were screened in three goat breeds (Barki, Damascus and Zaraibi) raised in Egypt. Polymerase chain reaction amplification followed by restriction fragment length polymorphism (PCR-RFLP) and DNA sequencing analyses, were used in order to differentiate these breeds. Both GH1-HaeIII/RFLP and IGFBP-3-XspI/RFLP showed only two genotypes (AB and BB) in all breeds, while AA genotype was absent. Regarding to GH1, both Barki and Zaraibi exhibited the highest frequency of AB genotype (0.95). For IGFBP-3, the highest frequency of AB was found in Barki (0.80). GH2-HaeIII/RFLP produced only two homozygous genotypes AA in Damascus and BB in both Barki and Zaraibi. The AB genotype was absent in the three breeds. In case of GH6 and MSTN-HaeIII/RFLP, the genotype BB was detected in all breeds, while AA and AB were absent. However, the sequence analysis of the PCR amplified fragment via BLAST indicated different SNPs among the three studied breeds. Whilst, both IGF-I–HaeIII/RFLP and PRL–Eco24I/RFLP revealed three genotypes (AA, AB and BB) with the highest frequency of AA in both Damascus and Zaraibi (1.00) for IGF-I gene. For PRL gene, the highest frequency of AB was found in Zaraibi (0.90). Finally, BMP-15 gene was monomorphic in all breeds when digested with HinfI. These results demonstrated that genes; GH1, GH2, IGF-I, IGFBP-3 and PRL were polymorphic. These polymorphisms may be utilized as effective markers for genetic differentiation between goat breeds. Moreover, different SNPs detected via sequencing of monomorphic genes; GH6 and MSTN could be recommended as potential markers, helping goat breeders in selection of goats for high growth rates.
