Abstract

Objectives: aim of this study is to investigate the efficacy of a combination of HA and BM-MSCs in accelerating wound healing after surgeries in rats receiving radiotherapy.
Materials & Methods: Sixty-four male albino rats were divided into four equal groups A, B, C and D after exposure to radiation. Full thickness wound was created on the back of the rats, three groups were injected subcutaneously; group A with stem cells, group B with HA and stem cells, group C: HA and group D was control group. Rats sacrificed on 3rd, 7th, 14th and 20th days after surgery. Wound healing was assessed by mRNA expression of genes interleukin-6, interleukin-1β and transforming growth factor-β (IL-6, IL-1β & TGF-β) by qRT-PCR assessment and the evaluation of collagen fibers formation.
Results: The healed tissues using BM-MSCs with HA showed the least number of inflammatory cells, the most well organized mature collagen fibers without scar formation and the better healing after assessment of changes in the gene expression of TGFβ-1 during mRNA analysis
Conclusion: Accelerated wound healing could be achieved by the use of HA and/or BM-MSCs in irradiated rats. The use of HA as a scaffold with seeding of BM-MSCs proved to be better in acceleration of wound healing as compared to the use of HA or BM-MSCs alone.
