Abstract

This thesis is devoted to the development of new analytical methods for the determination of five α-blockers; namely Terazosin hydrochloride (TER), alfuzosin hydrochloride (ALF), tamsulosin hydrochloride (TAM), doxazosin mesylate (DOX) and silodosin (SIL) in pharmaceutical preparations and/or biological fluids. The thesis is also concerned with the analysis of vardenafil, a phosphodiestrase type 5 inhibitor which is co-administered with alfuzosin and tamsulosin. The thesis comprises four main parts. Part I is a general introduction about the studied drugs including : pharmacological actions, chemical structures, physical properties, and literature review. Part II is devoted to develop simple, highly sensitive, specific and accurate RP-HPLC method for determination of ALF and TAM simultaneously with a co-adminestered drug vardenafil (VAR) in pure form, pharmaceutical preparations and spiked human plasma. Part III represents a novel, sensitive, simple and rapid RP-HPLC method for simultaneous determination of TER, ALF, TAM and DOX in pure form and pharmaceuticals. Part IV provides a novel, accurate, simple and rapid MEKC method for simultaneous estimation of TER, ALF, TAM, DOX and SIL. The method was successfully applied to their pharmaceutical formulations, content Uniformity and spiked human plasma. This thesis consists of four parts including 29 tables, 12 figures, 124 references and ends with an Arabic summary.
