Abstract

In this study a total of 2740 dairy cattle from Suez Canal Area were subjected to tuberculin skin test, 100 (3.6%) cattle were react positively, 85% of slaughtered tuberculin positive cattle showed visible lesions on while the other 15% showed no visible lesions. The microscopical examination of 100 slaughtered tuberculin positive cattle by direct smear, stained by Ziehl-Nelseen stain revealed the detection of the acid fast bacilli in 70 examined cattle samples with a percentage of (70%). While the bacteriological examination of 100 slaughtered tuberculin positive cattle by conventional culture method revealed that 78 cattle were positive for bovine tuberculosis with a percentage of (78%). The physical and biochemical identification of 78 M.bovis isolates. All of these isolates gave smooth colonies without pigmentation in dark or light at 37°C. And niacin production negative, while urease test positive. Regarding to comparison between tuberculin test and ELISA technique using PPD-B and commercial antigen mixture. Out of 100 tuberculin positive cattle, 89 serum samples collected from tuberculin positive reactors at slaughterhouse before being slaughtered were positive when used PPD as coating antigen. While 92 serum samples were positive when used commercial antigen mixture as coating antigen. So the sensitivity of commercial antigen mixture higher than the sensitivity of PPD-B. By IFN-Ɣ assay, 96 whole blood samples from tuberculin positive cattle before being slaughtered at slaughterhouse were positive gamma interferon with percentage 96%. While in contact cattle, out of 50 whole blood samples of tuberculin -ve reactors, 3cattle gave positive results with percentage (6%). These results showed that the sensitivity of IFN-Ɣ assay higher than the sensitivity of Microscopical examination, Culture technique, ELISA technique and tuberculin test. By real time PCR; out of 100 processed tissue sample of tuberculin positive animals, 96 tissue samples were positive with percentage (96%). While all tested isolates of mycobacterium spp. were positive with percentage (100%) by using the primers of mycobacterium tuberculosis complex.
