Abstract

The present studies were divided in two parts : Part I: This part was carried out at the Department of Surgery, Anesthesiology & Radiology - Faculty of Veterinary Medicine - Zagazig University, Egypt. The experiments, animal handling and surgical protocols were conducted according to the guidelines for animal care and use committee of Zagazig University, Egypt, and approved by the Ethics Committee at the Faculty of Veterinary Medicine – Zagazig University. It was objective to evaluate the efficacy of CSH & DB mixture in repair of induced 5mm full-thickness segmental defect in the mid-diaphysis of the femur in dogs.
Part II: This part was carried out at the Key Laboratory of Oral Biomedicine Ministry of Education, School & Hospital of Stomatology, Wuhan University, People’s Republic of China. The experiments, animal handling and surgical protocols were conducted according to the guidelines for animal care and use committee of Wuhan University, People’s Republic of China, and approved by the Ethics Committee at the School of Dentistry. It was objective to: (i) Evaluate the osteo-inductive potential of rhBMP9 for bone regeneration in rats, (ii) Investigate a novel NIR-triggered drug delivery platform loaded with black phosphorus (BPs) and strontium chloride (SrCl2) for bone regeneration in rats, and (iii) Compare the efficacy of PRP and i-PRF on regeneration of critical-sized osteochondral defects in the trochlea of the femur in rabbits. In part I, twenty seven (27) adult apparently healthy male mongrel dogs were subjected to an induced 5mm full thickness segmental defect in the mid diaphysis of the femur of the right hind limb to evaluate the osteogenic potential of CSH & DB mixture. These animals were divided into three groups (n=9 in each group) : group I: (control group) in which the femur defects were left empty, group II : (sham group) in which the femur defects were wrapped with polypropylene mesh, and group III: (experimental group) in which the femur defects were implanted with CSH & DB mixture and wrapped with polypropylene mesh. All animals were investigated clinically to determine the score of lameness along the study period (24 weeks). All animals were euthanized at 6, 12, and 24 weeks after surgery for radiological, computed tomography, gross and histological examinations.
