Abstract

The present investigation was conducted to the work done at the Tissue Culture Laboratory, Department Vegetables, Faculty of Agriculture, Assiut University, Assiut, Egypt during the period 2014 - 2016.This work aimed to reduce effectively or eliminate the media contamination occurred during the plant tissue culture process on explants and medium. This was done through using new methods of explant sterilization and un-autoclaved self-sterilized medium that can be used to grow plants such as strawberry plants.>Data on contamination severity and proliferation speed presented showed highly significant differences among sterilization materials at different concentrations with non-significantly effects between varieties of strawberry under study. Dettol® at 100 ml/L treatment recorded the lowest values of contamination percentage on the explants (0.46 %) compared to the control treatment which recorded (28%) contamination of explants. On the other hand, control treatment and all concentrations of Clorox® recorded a speedily proliferation (1, 2.2, 2.3 and 2.9 days respectively) more than the other treatments. At the same time, the strawberry explants which treated with 100 ml/L Dettol® had damaged.
Contamination percentage in tissue culture techniques considered as the most economical part of its production. Data on contamination severity presented showed highly significant differences among Assiutmix-3 levels as anti-contamination compound on the strawberry varieties. Assiutmix-3 at (4 m/L) were the best treatment for decreasing the medium contamination (0.40%) rather than the other concentrations and the control treatment (un-
Summary- 36 ->treated) which recoded (99.5%). Strawberry cultivars had no significant effect of Assiutmix-3 concentrations.
As well as, the results of investigation showed significant effect on anti-contamination compound cultivars and their interaction adding Assiutmix-3 with 4ml/L to the culture media gave the highest multiplication rate, shoot length, average weight of shoot, number of leaves/shoot, number of root, root length and average weight of root/shoot, while adding Assiutmix-2 with 1.5 ml/L to the culture media gave the highest value of shoot length.Festivelle cultivar was better than Americana cultivar in all studied traits.
