Abstract

Isolation of mesenchymal stem cells (MSCs) from full term umbilical cord blood (UCB) may appear easy due to plenty of cord blood and easily isolation using low cost ficoll hypaque solution, but it is a time-consuming process and results in a low yield of cells. Preparation of ovarian cancer cell line from the tumour itself has the advantage to isolate cells from early and late stages, also, from the primary site or area of metastasis, unlike the preparation from ascites that is formed in more advanced stages. MTT assay results demonstrate an inverse relationship between cell viability and concentration of mesenchymal stem cell extract. There was an increase in early apoptotic cell populations and late from untreated to treated cells. The increase in apoptotic cells was also reflected by changes in the light scatter properties for the untreated and treated tubes (FSC-A vs SSC-A plots). The increased peaks in the sub-G1 phase and positive annexin VFITC staining were suggestive that the conditioned media anti cancer effect was via the apoptotic mechanism. The expression of embryonic stem cells markers (SOX2, OCT4 and NANOG) is crucial for progression of ovarian malignancies. The expression levels of SOX2, OCT4 and NANOG were affected by conditioned media in concentration related manner compared with control. Although this study proved the suppresive effect, Multiple experiements are needed to identify the key substance, and further trials to delineate the effect in vivo.
