Abstract
The present study was designed to investigate the adverse effect of Nitazoxanide and protective effect of Vitamin E and to show the result of the combination between Nitazoxanide and Vit.E on hepatic and renal disturbance induced in rats by Nitazoxanide.
Experimental design: After one week of acclimatization, eighty female albino rats randomly allocated into 4 equal groups, each of 20 rats.
The results were summarized as follow:
1-Effect of Nitazoxanide, Vit.E and their combination on hematological parameters (HBG, RBCS, HCT, WBCS, Platelets count). Oral administration of Nitazoxanide (18mg/kg b.wt once daily for 14 days) showed a significant decrease in HBG, RBCS, HCT, WBCS, Platelets count compared with control group. Administration of Vit. E and it,s combination with Nitazoxanide showed increase in HBG, RBCS, HCT, WBCS, Platelets count compared with Nitazoxanide group.
2-Effect of Nitazoxanide, Vit.E and their combination on liver parameters (ALT, AST, ALP, total serum proteins, serum albumin, total serum bilirubin).
Oral administration of Nitazoxanide (18mg/kg b.wt once daily for 14 days) showed a significant increase in (ALT, AST, ALP) while total serum proteins, serum albumin, showed a significant decrease and a non- significant change in total bilirubin compared with control group. Administration of Vit.E and its combination with Nitazoxanide showed a decrease in (ALT, AST, ALP) while total serum proteins, serum albumin, showed an increase and a non- significant change in total bilirubin compared with Nitazoxanide group.
1-Effect of Nitazoxanide, Vit.E and their combination on kidney parameters (creatinine and urea). Showed a significant increase in (creatinine and urea).
Administration of Vit.E (18mg/kg b.wt once daily for 14 days) and its combination with Nitazoxanide (18mg/kg b.wt once daily for 14 days) showed a decrease in(creatinine and urea) compared with Nitazoxanide group.
2-Effect of Nitazoxanide and Vit.E on antioxidant activities: A] CAT, SOD and GPX activity: Showed a significant decrease in CAT, SOD and GPX activity compared with control group. Showed a significant increase in CAT, SOD and GPX activity. B] Lipid peroxidation (MDA): Showed a significant increase in MDA activity compared with control group. Administration of Vit.E showed a significant decrease in MDA activity.
3-Histopathological findings: According to the liver, Oral administration of Nitazoxanide showed markedly dilated congested central vein surrounded by rows and cords of hepatocytes with clean cytoplasm due to severe fatty change in the 7th day and dilated congested central vein with mild fatty change of the hepatocyte in the 14th day.
