Abstract

Oral squamous cell carcinoma (OSCC) is one of the most common and lethal head and neck malignancies , OSCC is now a global health problem with increasing incidence and mortality rates. The major environmental risk factors responsible for the development of OSCC include betel nut chewing, cigarette smoking, alcohol consumption, and exposure to high-risk human papillomavirus.
Despite increased experience in therapy, including improvements in surgical technology and adjuvant treatments, the overall outcome of OSCC has not improved, indicating an urgent need for a new strategy for OSCC treatment.
In this study, we used quercetin which is one of the most abundant dietary flavonoids. It is found in onions, apples, green tea, grapes, and berries and we investigate its cytotoxicity effect on HEP-2 cell line using MTT assay and morphometric analysis. The change in cell cycle distribution of HEP-2 cells was detected using flow cytometry.
The results showed that quercetin has a cytotoxic effect on the viability of HEP-2 cells using MTT assay. The morphometric analysis confirmed the results of MTT assay as there was a decrease in the nuclear area factor(NAF) in a dose dependent manner.
The flow cytometric analysis of Quercetin shows pre G1 apoptosis with cell cycle arrest at G1/S phase.
