Abstract

This study aimed to evaluate the effect of caloric restrictions and L-carnitine on antioxidant status and the expression of genes associated with lipid metabolism in rats.
Forty adult male albino rats weighed (14-150 gm) were used in this study and were divided to 4 groups. In the first group (control group), in second group rats were supplemented with L-carnitine 100 mg /kg body weight. In group three rats fed on caloric restricted diet 30-40% caloric restricted diet ,group four rats fed on caloric restricted diet and supplemented with L-carnitine which given at a dose of 100 mg/kg body weight orally to the rats
The result of this study showed that the activities of SOD, GSH in liver tissue homogenate, and total antioxidant capacity in serum were increased in group three, and four. The level of cholesterol triacylglycerol and LDL-C in serum was decreased in group three and four, while it was increased in control group. The levels of oxidative stress marker MDA in liver tissue and serum were decreased caloric restricted groups than control group. Thyroid profile and testosterone were decreased in caloric restricted group in comparison with control groups and there was increase in cortisol hormone in caloric restricted groups in comparison with control group. The expression of Leptin and HMG-CoA reductase were down-regulated in group three and four, while it was upregulated in group one. The expression of Adiponectine was up-regulated in group three and four, while it was down-regulated in group one.The histopathological examination showed improved architecture of liver in caloric restriction group than control group and by immunohistochemistry SOD activity was increased in caloric restriction groups three and four than control group.
