Abstract

Biofuel production from photosynthetic bacteria is the main target of the presented work. Two groups of photosynthetic bacteria; purple sulfur (PSB) and purple non sulfur (PNSB) were isolated from different localities in Egypt. Nineteen samples werecollected from different localities in Egypt;Assiut, Alexandria, Aswan, El Bahr Al Ahmar and North Sinai governorates. These samples were inoculated in RÄH& Pfennig’s media and purified by shake dilution method. SevenPNSB and 3PSB wererecognized and identified according to Bergy’s Manual. Morphologically, one important characteristic of the purple bacteria is the reddish pigmentation which indicated the presence of the photosynthetic pigments (carotenoids and bacteriochlorophyll-a). Purple sulfurbacteria differ from PNSB in the ability to form food reserve as sulfur and poly hydroxybutayrate granules inside the cell. Physical characterization of PHB include Transmission Electron microscopy (TEM) image and Fourier transformed infrared spectroscopy spectrum (FTIR) proved the nature and amount of this molecule. All isolates (PSB and PNSB) were tested for nitrogenaseactivity and hydrogen production then;five isolates were selected forfurther investigation. The most nitrogenase activity and hydrogen production isolates were RhodobactersphaeroidesMG 588088;AllochromatiumvinosumMH973736 for PNSB and PSB; respectively.
