Abstract

The present study was performed to study the role of Cytochrome P450 1B1 (CYP1B1) expression in inflammatory cells on obesity gene expression in hepatocytes of obese mice. Part I:male mice were divided into 5 groups and fed HFD for 11 weeks. The groups were divided as follows; group I: Control group: WT mice fed HFD group II: Cyp1b1-ko (KO) on HFD: cyp1b1 from all cells of the body. group III: C-Cre occurs in all tissues, including germ cells.group IV: L-ce HFD Cyp1b1 deletion occurs iinflammatory cels. group V: C-Ce on HFD Cyp1b1 deletion in other cells At the end of the experimental period (11 weeks), mice were weighed, and blood samples were collected by cardiac puncture from all these mice and used for determination of the some parameters: PCR was done to measure the expression. Part II: mice aged (5-7 weeks) were divided. BM cells were obtained from the femurs of all mice. BM cells were treated with 20 ng/ml M-CSF. On day 6, the media changed with (IFN-γ) 20 ng/ml ( for 3,6,12 and 24 hours. At the end of each time point, cells were collected to measure gene expression using PCR. The results could be summarized as follows: general deletion of cyp1b1 caused significant decrease of body weight, epididymal fat pad weight and liver weight. It significantly affected the hepatic gene expression and showed the same effect on macrophages gene expression as it significantly decreased the expression of M1 markers while it didn’t affect the expression of M2 markers. deletion of cyp1b1 in inflammatory cells produed some of the same effects as deletion of cyp1b1 in all cells while deletion in all cells caused an opposite effect to general deletion . from these results, we can conclude that deletion of cyp1b1, in case of diet induced obesity, could significantly reduce body weight and improve the metabolic state which could be attributed to change in macrophages polarization by switching from M1 to anti-inflammatory M2 phenotype and also due to significant changes in hepatic gene expression concerned with energy utilization.
