Abstract This study was conducted on 60 rats (120-170 gram). After 2 weeks of acclimatization, Rats were divided according to diet as follow: normal diet rats and high fat diet rats. The rats fed HFD for 2 weeks then rats fasted for 16-18 hrs. With free access to water (ad-libitum) before induction of diabetes by a single I/P injection of STZ by the dose of (35mg/kg). Diabetes was confirmed by estimation of fasting blood glucose level 72 hrs. post injection by using blood glucose test strips. The blood glucose level higher than 200 mg /dl means diabetic. Then the normal diet rats divided into two groups served as :control, (VD) rats treated with vitamin D (1000 IU/ kg B.W I/M day after day) and the confirmed diabetic rats divided into 4 groups (DM)diabetic untreated group,(DM+VD) diabetic treated group with vitamin D (1000 IU/ kg B.W I/M day after day),(DM+ Met) diabetic treated group with metformin (100 mg / kg B.W orally/ day),(DM+ VD+ Met) diabetic treated group with both vitamin D (1000 IU/ kg B.W I/M day after day) and metformin (100 mg/kg B.W orally/ day).The treatment duration is for 4 successive weeks for all groups. After 4th weeks of treatment, 5 rats were picked up randomly from each group and blood samples were collected from medial canthus of the eye to investigate some hematological and serum biochemical parameters . our study confirm best treatment for the combined group and confirm this by histopathology for pancrease ,liver,kidney,spleep. |
