Abstract

Specific Aim : this study was designed to evaluate, in vitro, different therapeutic properties (biocompatibility, anti-inflammatory and antimicrobial ) of Oeanolic acid for use in root canal treatment. Owing to the potential side effects of the different materials used in root canal treatment. So that, obtaining excellent economical, biocompatible material that would be able to totally eliminate bacteria from root canal system and act as an excellent analgesic and anti-inflammatory to control the postoperative pain. Materials and Methods : In this study, the herbal medicinal extract used for the experimental purpose was Oleanolicacid . WI-38 cell line was used to evaluate the biocompatibility of Oleanolic acid by MTT cytotoxicity assay in comparison to the most commonly used irrigations and medications in endodontics [2.5% Sodium hypochlorite NaOCL, 2%Chlorhexidine (CHX) solution, Calcium hydroxide Paste ,Chlorhexidine CHX gel ,Mix of Ca(OH)2 / CHX, Ledermix, Triple antibiotic paste (TAP)and Double antibiotic paste(DAP)]. Cyclooxygenase Inhibitory assay as well as Lipoxygenase inhibitory assay were used to evaluate anti-inflammatory activity of Oleanolic acid. Also Autoantibody Profiling System (Elisa Kits) for measuring IL-6 and TNFα was used to evaluate anti-inflammatory activity of Oleanolic extract.. The antimicrobial evaluation of Oleanolic acid against the most resistance microorganisms in root canal treatment (E. faecalis and candida albicans) was performed using microdilution test against gram positive bacteria Enterococcus Feacalis and candida albicansfor measuring the minimum inhibitory concentration (MIC) to determine the effective antimicrobial concentration. Results : The biocompatibility of Oleanolic extract was to be compared to the all tested root canal irrigations and medications, as indicated by MTT assay showing good biocombatibility of Oleanolic Acid. The result of anti-inflammatory assays showed that, for both Cyclooxygenase and Lipoxygenase assays, Oleanolic acid had higher anti-inflammatory action than that of ledermix. Comparable results were found withIL-6 and TNF α showing that Oleanolic acid either alone or when added to LPS always had a lower level ofIL-6 and TNF expression than that showed with LPS alone indicating good anti-inflammatory action of Oleanolic acid. Broth microdilution test showed that, the minimal inhibitory concentration of Oleanolic acid against each E. fecalies as well as C.albicans was 6 g/ml for each. Conclusion : within the limitation of this study, it was found that Oleanolic acid could be considered to have good potential as a root canal irrigant or medicament. The commonly used irrigations and medications should be used with caution in endodontic theraby, since the drugs act not only against microorganism ,but also on cells of the periradicular tissue. Recommendations : Further laboratory and clinical investigation should be carried out to validate finding beneficial of Oleanolic Acid as intracanal medicament or as any other endodontic materials.
