Abstract

This work is based on two principal points, the first was to choose some genera of different medicinal plants and collection of some human pathogenic bacteria and fungi in addition to citrrus brown. The second point was to evaluate the and screen the antimicrobial, antiparasitic and anticancer activities of these medicinal plants. The most important results can be summarized as the following points:
1-Preparation of extracts of 12 medicinal plants using different solvent systems like water, methanol and ethanol
2-Collection of some pathogenic bacteria from KSA and Egypt, purification and identification of these pathogens by different.
3- Results of identification showed the presence and prevalence of S.aureus, E.Coli,P.aeurigonoa, S.typhi and C.albicans.
4- Qualitative assay was performed using well plate method to screen and detect the antimicrobial activity and results assessed that only 4 plants showed the antimicrobial activity, these are rosemary, black cumin, ginger and myrrh.
5-quantitave assay followed the qualitative one to determine the efficacy and potency of each extract, and results showed that methanol extract of each plant nearly was the most efficient one and also, rosemary methanol extract was the most efficient one recording 19.0 mm inhibition zonein case of E.coil followed by rosemary ethanol extract , myrrh, ginger then black seed.
6- GC-MS analysis was carried for the plants methanol extracts to determine their major component and it showed presence of important substances like oils and phenols.
7-Throbolytic effect of all extracts types for the 4 plants was screened and they proved that they are potent antithrombotic agent that dissolved plasma clots in vitro
8-Cytotoxic effect of these plants was detected in addition to cell viability using their methanol extracts against human colon cancer cell line and the results showed that they were candidates for cancer treatment via decreasing the viability of cancer cells, the most efficient plant was myrrh recording IC50 8.24µglml
9-Gene expression study was also performed on 3 cellular genes responsible for regulation of cellular activities and cell apoptosis, these are BAX, BCL2 and P53 genes and results showed that treatment yielded up regulation of both p53 and BAX and down regulation of BCL2 which resulted in induction of apoptotic response of cancer cell making these substances promising anticancer agents
10- Mode of action of rosemary methanol extract was carried out via protein leakage test in addition to TEM and SEM imaging, results showed its efficacy in inducing protein leakage from bacteria cells recording the highest effect against S.aureus. In addition, SEM and TEM revealed that bacteria treated with rosemary methanol extract showed distortions and abnormal cell structure
11- Antiparasitic effect of the plants aqueous and alcoholic extracts was detected against the citrus brown mite and results showed that both extracts showed inhibitory effects on egg deposition, egg hatching and reproduction of this parasite. Moreover, methanol extracts were superior to the aqueous extracts. So, these substances can behave as bioacaricide as they reduced the number of eggs laid, eggs hatchability and reproduction potential of E.orientalis.
