Abstract

Norfloxacin is a synthetic second generation antimicrobial agent of the fluoroquinolones class. The mode of action of norfloxacin is depending on inhibition of DNA gyrase, which essential for supercoiling of the bacterial chromosomes. Ibuprofen is a propionic acid and one of the non-steroidal anti-inflammatory drugs (NSADS) that has both analgesic and antipyretic effects. Ibuprofen is used for short-term treatment of pain, inflammation from injuriesThis study was designed to explore the possible effects, if any, of norfloxacin and Ibuprofen on male rats through examination Blood picture of treated rat , Serum total testosterone level , Sperm picture, The testes and epididymal tissues (Histopathological examination). Sixty clinically healthy mature male rats (200-300 g) were classified to four equal groups (15 rats ofeach) as the following: First group (control): administered 0.3 ml saline by stomach tube for ten successive days Second group: orally administered norfloxacin (500 mg/kg b. wt) by stomach tube for 10 successive days. Third group: orally administered ibuprofen (30 mg/kg b. wt) by stomach tube for 10 successive daysFourth group: orally administered norfloxacin (500 mg/kg b. wt) and ibuprofen (30 mg/kg b. wt) by stomach tube for 10 successive daysFive blood samples were collected from each group in the first day , the first week then , the second were collected after administration of drug from the heart . Half the sample in test tube contain EDTA , to evaluate blood picture , the second half in test tube without EDTA to be clot at the room temperature . the clotted blood samples were centrifuged for 15 minutes at 3000 r.p.m to obtain a clear serum . Epididymal sperm content were collected by diffusion method , in which cauda epididymis is placed in petri-dish having an aliquot of physiological buffered saline solution . The recorded result evoked non – significant alteration in total erythrocytes count of all treated group at first day , first week and second weak and 2 weeks with treatment compared with control Norfloxacin in group 2 or with ibuprofen in group 4 cause a significant increase in PCV in first day , first week and second week , the result show a significant decrease in MCHC with group treated with norfloxacinThe result illustrated some abnormal alteration in the sperms picture of mature male rats given the tested drugs Effect of the tested drugs on total serum testosterone level showed regular cross section of seminiferous tubuels lined with full thickness of spermatocytes and epididymis lesions We recommended that neither folxacin nor ibuprofen should be used together during the treatment of males for long period because of their negative effect on fertility in males.
