Abstract

ALA (1,2dithiolane-pentanoic acid) is a naturally occurring cellular antioxidant, which have potent antioxidant property and can act as free radical scavenger of ROS and reactive nitrogen oxide species (RNOS). Diabetes Mellitus (DM) would result from a defect in insulin secretion and /or activity that cause different disorders in metabolism which characterized by diabetic hyperglycemia. The current study was conducted to evaluate the protective role of ALA on rats exposed to streptozotocin and secondarily complicated with nephropathy through examination of Fasting serum glucose, Fasting serum insulin, Serum creatinine, Serum urea, Malondialdehyde (MDA) concentration, Nitric oxide (NO) concentration, Glutathione reduced (GSH) concentration, Glutathione peroxidase (GPX) activity it, Catalase (CAT) activity, Caspase 3 and Histopathological examination of kidney tissue. Forty eight male albino rats (100-125g) were divided into four equal groups: 1- group I(control):was fed on normal commercial diet. 2- group II(ALA group):was fed on ALA rich diet (20mg/kg diet). 3- group III(STZ group):injected with STZ (35mg/kg b. wt) i.p for induction of DM type 2 and was fed on normal die. 4- group IV(STZ+ALA): injected with STZ (35mg/kg b. wt) ,was fed on ALA rich diet. At end of experiment blood samples were collected and kept to clot at RT then centrifuged at 3000 rpm for 15 min then separated to obtain serum for determination of (serum glucose, insulin, creatinine and urea) then animals were dissected to obtain kidneys to be homogenized and used in (Oxidative stress and antioxidant markers, apoptosis marker as caspase 3) and the other part used for histopathological examination The results of the current study revealed that ALA alleviate the oxidative damage induced by streptozotocin exposed rats and improving antioxidant status (catalase, reduced glutathione), decreasing oxidative and nitrosative stress markers(malondialdehyde and nitric oxide), improving renal damage markers. Enhancing glycemic status and decreasing caspase 3 concentration.
