Abstract The sponge material used in this work was collected by Prof. Dr. Safwat Ahmed (Suez Canal University) from Sharm El- Shaikh at the Egyptian Red Sea, air-dried and stored at low temperature (-24 ºC) until processed. 1-Material for Phytochemical and biological Study: The freeze-dried sponge material (10 g) was extracted with methanol–methylene chloride (1:1). The resulting crude extract was initially partitioned between H2O and pet-ether yielding pet-ether fraction, followed by dichloromethane, ethyl acetate and butanol The organic phase in each step was separately concentrated under vacuum, yielding pet-ether fraction (1g), dichloromethane (250 mg), ethyl acetate (1 g) and butanol (1.5 g) The preliminary phytochemical screening of the Freeze -dried sponge Amphimedon sp. revealed: 1. The dried sponge Amphimedon sp. contains carbohydrates / or glycosides, sterols, alkaloids and/or nitrogenous compounds and /or triterpenes. 2. Absence of saponins, cardenolides, tannins, Flavonoids. 1-Extraction and fractionation. A) Preparation of the methanolic-methelyene chloride extract 2- Fractionation of the methanolic- methelyene chloride extract 2- Isolation of the main constituents of the sponge Amphimedon sp. 1-Column chromatographic fractionation and isolation of the pet-ether fraction 2-Column chromatographic fractionation of the ethyl acetate fraction. We carried out metabolomic analysis and derplicated and identified 90 compounds. Compounds identified and dereplicated from high resolution mass spectral data sets from fractions of of the marine sponge Amphimedon sp. by utilizing macros and algorithms that coupled MZmine with both in-house and commercial databases speciallay Marinlite data base. The compounds were listed in the Table (31). from the previous identified dereplicated compound we can concluded that Amphimedon sp. is a very rich sponge, rich with bioactive compound with diverse classes and diverse biological activity. The high number of unidentified compounds highlights the chemical potential of this sponge as a source of new metabolites. |
