Abstract The pathogenesis of acute gastric mucosal injury are not yet fully elucidated. It may be caused by the imbalance between protective factors and destructive factors. The main protective factors include adequate mucosal blood flow, mucosal bicarbonate barrier, endothelial cell regeneration and prostaglandins. On the other hand, destructive factors include gastric acid, pepsin, bile, reperfusion injury and oxygen free radicals. When the aggressive factors overwhelm protective factors, this unbalance can lead to the generation of ulcers. Indomethacin, a widely-used non-steroidal anti-inflammatory drug, inhibits both COX isoforms and thereby exacerbates damage to gastric tissue and leading to induction of oxidative stress, modulation of the NO system and reduction of gastric mucosal blood flow.
