Abstract

Hyperlipidemia can be severely detrimental as a major risk factor for fatty liver disease, atherosclerosis and cardio-cerebrovascular disease. This study goaled to explore the ameliorative effect of ethanolic extract of Flaxseed and Garden cress seeds in Hyperlipidemic and NAFLD rat model. Rats classified into six groups (1): control rats were fed on standard diet. Groups (2,3,4) rats fed on high fat diet (HFD) for 12 weeks. Groups (3,5): rats daily supplemented ethanolic extract of flaxseed seeds (EEFS) (400mg/kg b.w) per- os starting from seventh week till end of experiment. Groups (4,6):rats orally received ethanolic extract of garden cress (EEGS) (400mg/kg b.w/day) beginning from the seventh week till the end of the experiment. HFD- fed rats evoked an increment of body weight gain, hyperlipidemia, serum liver enzymes leakage, elevation in serum cardiac enzymes, enhanced inflammatory burden. Additionally, HFD motivated hepatic oxidative stress, lipid peroxidation. NAFLD and cardiac damage that confirmed with histopathological findings. Meanwhile, EEFS or EEGS supplementation after HFD feeding attenuated hyperlipidemia as expressed with lower TC, TAG, LDL-c in line with down-regulation of HMGCR and VEGF expression than HFD group. As well as EEFS or EEGS treatment in HFD group enriched the hepatic antioxidant via elevated GSH content, SOD, CAT activities along with lessened MDA and nitric oxide level. And also, evoked anti-inflammatory prospect. In vitro, EEFS and EEGS displayed antioxidant properties via DPPH radical scavenging. Additionally, HPLC screening realized high phenolic, flavonoids, vitamin C content in both extracts. Our data achieved that, daily administration of EEFS or EEGS at dose of 400mg/kg b.w for six weeks is safe and possess hypolipidemic, anti-obesity, hepatoprotective, cardioprotective, antioxidant , anti-inflammatory, anti-atherogenic, anti-angiogenic effects in HFD-induced hyperlipidemic and NAFLD. Hence, EEFS and EEGS could be safely applied in atherogenesis- induced hepatic and cardiac disease management
