Abstract

This study was carried out to investigate the beneficial effect of vitamin D -rich diet on diabetic rats. Rats were divided into 4 groups: group I, received control diet, group II, received HFD diet for 2 weeks followed by two doses STZ 30 mg/kg Bwt at an 24 hrs interval, then recieved with control diet till end of experiment which was kept as a diabetic group , group III, received HFD diet for 2 weeks followed by two doses STZ 30 mg/kg Bwt at an 24 hrs interval, followed by dose of vitamin D(3300 IU/Kg diet) for another 3weeks which was kept as a treated group, group IV, received vitamin D for one week followed by HFD diet for 2 weeks followed by two doses STZ 30 mg/kg Bwt at an 24 hrs interval, followed by dose of vitamin D (3300 IU/Kg diet) for another 2weeks which was kept as a preventive group. Blood and tissue samples were collected after six weeks for measurement of serum glucose, insulin, HOMA IR, 1,25 OH vit D, PTH, serum lipid profile: total cholesterol (TC), triacylglycerol (TAG), HDL-cholesterol (HDL-C), LDL- cholesterol (LDL-C), VLDL-cholesterol (VLDL-C); blood and tissue antioxidants :reduced glutathione (GSH), superoxide dismutase (SOD) and catalase; oxidative stress parameters, serum lipid peroxidation product: malondialdehyde (MDA) and histopatholgical examination of brain, pancrease and liver; Acetylcholine Esterase (AChE), Amyloid Beta Precursor Protein (AβPP) gene expression and tissue caspase 3, TGF . from these results it can be concluded that dietary supplementation of vitamin D specially as a preventive measures ameliorate the neurodegenerative effect of T2DM by increasing brain antioxidant activities and inhibiting amyloid beta protein plaques formation and vitamin D assured great protection against complication of type II diabetes in rats. It can be recommended to reduce the risk of susceptibility for diabetes as it improve lipid profile, antioxidant status and Amyloid Beta Precursor Protein and Acetylcholine Estrase gene expression in normal individuals.
