Abstract

Our attention of the current study was directed to use ZnONPs in the treatment of type 2 diabetes in Streptozotocin – induced diabetic rats, evaluate its effect on blood glucose level and carry out the following investigations to fulfill the following objectives: 1. Study the effect of Zinc oxides nanoparticle in the modulation of oxidative stress associated with diabetes mellitus 2. Determination the mechanism of the action of Zinc oxides nanoparticle through the evaluation of the gene expression of glucose transporter GLUT-4; Tumor necrosis factor (TNF); Interleukin 6 (IL-6); CD4+ and CD8+ T cells. 3. As well as studying the histopathological changes of pancreas tissues associated with diabetes mellitus before and after treatment with ZnONPs. It can be concluded from the current results that treatment of diabetic rats with zinc oxide nanoparticles (10 mg / kg body weight) showed a marked improvement in the effects of Streptozotocin (STZ), which led to a decrease in the level of blood glucose and glycosylated hemoglobin and an increase in insulin levels, as well as reducing oxidative stress through stimulation the activity of superoxide dismutase (SOD) and inhibition the activity of glutathione reductase (GPX). It also inhibited the production of pro-inflammatory markers such as IL6, TNF-α, as well as the level of CD4 and CD8. On the other hand, the treatment of diabetic rats with zinc oxide nanoparticles (10 mg / kg of body weight) resulted in elevation of gene expression of GLUT 4 gene and restore the pancreatic cells to produce the insulin.
