Abstract

Objective: To evaluate the effect of platelet rich plasma in combination with bone marrow mesenchymal stem cells in albino rats’ periodontal regeneration. Methods: Eighty-four adult female rats were selected for this study; four of them were used for PRP preparation, three for stem cell isolation, and one for investigation of normal histology. Ligatures were placed subgingivally around the lower incisors of 76 rats, removed after 14 days, and one rat was euthanized for examination of the periodontitis model. The remaining 75 rats were all subjected to careful scaling and root planning, and subsequently divided into 5 equal groups (n=15) to receive injection of the treatment as the following; group I (SRP only), group II (0.2ml PBS), group III (0.2ml PRP), group IV (1×106 cells BMMSCs in 0.2 mL PBS), and group V (1×106 cells BMMSCs in 0.2 mL PRP). Five rats of each experimental group were euthanized after 1,3,6 weeks, and the specimens were processed, then evaluated using H&E, Masson trichrome, Osteocalcin and digital morphometric assessment followed by one-way Anova statistical analysis. Results : group VI showed enhanced results regarding alveolar bone and PDL regeneration compared to all groups especially in the period of 6th week. group V revealed better results similar to group VI in the periods of 1st and 3rd weeks, but worse alveolar bone results after 6th weeks of treatment. Conclusion: Local injection of BMMSCS was successful in regeneration of periodontal tissues of rat incisors with induced periodontitis. Besides, combination of BMMSCs and PRP showed short-term enhanced results which requires further investigations regarding the dose and separate administration of both.
