Abstract This study is concerned with the effects of low doses of gamma radiation on some Egyptian varieties of faba bean at morphological, cytological, biochemical and molecular levels in the M1 and M2 generations. Seeds of six faba bean varieties (Sakha 1, Nubaria 2, Nubaria 3, Giza 3, Giza 716 and Giza 843) were exposed to 20, 40, 60, 80, 100 and 200 Gy of g-radiation. In the M1and M2 plants, the doses of 20, 40 and 60 Gy were the most effective in increasing shoot length, root length, shoot fresh weight, root fresh weight, shoot dry weight and root dry weight, leaf area, leaflet length, leaflet width, pod length, number of seeds per pod, number of pods per plant, number of seeds per plant, seed yield per plant and 100-seed weight. Mitotic activities in root meristems were enhanced by the doses of 20, 40 and 60 Gy of g-radiation, chromosomal abnormalities such as C-metaphase, stickiness, disturbed metaphase, lagging chromosomes, chromosome bridges, vagrant chromosomes, sticky anaphase and nuclear abnormalities as micronucleus and binucleate cells were induced. At the molecular level, ISSR pattern using seven primers revealed 81 bands, 75 bands were polymorphic and 6 bands were unique in M2 plants. The doses effect on M1 plants are transmitted to the M2 plants. Some effects of the applied doses increased progressively in the M2 plants supporting the use of g-radiation to produce new mutants. |
