Abstract

This study aimed to assess the Effect of Some Immuno-Stimulants on Immune Response of Infectious Bursal Disease Virus In Chickens. The present study was conducted on 55 clinical samples (bursa of fabricius, liver, spleen and kidney) which were collected from 55 chicken farms were suspected to be infected with IBDV. IBDV isolation from 55 collected samples onto chorioallantoic membrane of 11 days old ECEs was conducted and three serial passages were carried out for each sample. Evaluation of the effect of immunostimulants (G.I and/or P.A) on immune-response of expirementaly vvIBDV infected chicken which tested by the following parameter : mortality rate, total leucocytic count, lymphocytic count, ELISA to detect the antibody titer, bursa/body weight ratio and spleen/body weight ratio every 7 days from start of exiperment. Agarose gel electrophoresis of the conventional RT-PCR products obtained from amplification reaction of RNA extracted from third egg passage homogenates revealed the positive amplification of 743 bp product fragment of VP2 gene from 10 selected isolates. Phylogenic analysis of the nucleotid sequence of amplified IBDV-VP2 gene revealed that the isolate in this study (mans018) showed close relationship with other Egyptian serotype1, genogroup 3 vvIBDV strains from GenBank with 95.8-98.1 % identity. Expiremental study concluded that there was improvement in mortality rate in challenge groups especially in prophylactive then therapeutic groups (G11) in which both N.S and P.A were used before and after challenge with vvIBDV. TLC and LC were significant increased in groups supplemented with G.I and P.A and the higher percentage in challenge groups was found in prophylactive then therapeutic groups especially in G11 (G.I and P.A added before and after challenge with vvIBDV). ELISA revelead that the prophylactive then therapeutic groups (G9-G11) showed higher titer of antibody than other group didn’t took immunostimulants especially in (G11) where supplemented with both N.S and P.A before and after challenge with vvIBDV.
