Abstract

Nanovaccines have a great interest at the last years to enhance the absorption of biosafe subunit vaccine agents inside the body. This type of vaccines depends mainly on a nanoparticle platform that can potentiate the immune response in adjuvant-like manner by enhancing antigen uptake through antigen presenting cells and thus, stimulating a protective immune response against infections.
In this work, we used nanochitosan as a delivery platform that enhances the engulfment of bovine serum albumin by phagocytic cells and inducing strong systemic and mucosal immune response against immunogenic peptides involved inside its peptide sequence. The interaction between chitosan nanoparticles (CSNPs) and bovine serum albumin (BSA) has been investigated using spectroscopic techniques. Results showed that the Synthesized CSNPs were irregular in shape with size of ~ 42 ± 6 nm. BSA – CSNPs bio-complex (BIO-BSA-CSNPs) interaction depends mainly on the static quenching by the formation of ground state complex. This leads to a decrease in alkaline media and at physiological temperature by about 4 and 6 %, respectively. The safety and biocompatibility of BIO-BSA-CSNPs inside the body was also investigated after intraperitoneal injection (IP) of male mice for nine days. In vivo results revealed that free CSNPs and BIO-BSA-CSNPs didn’t change the body weight, the relative weight and histology of liver, spleen and kidney. This study exhibited non-significant changes in hematological and biochemical parameters with an observed hypocholesterolemic effect in all treated groups except BIO-BSA-CSNPs 2 treated group (group F) in compared to the negative control mice after nine days. Also, BIO-BSA-CSNPs 1 (group E) and 2 (group F) treated groups showed a significant increase of total WBC count after nine days of administration due to CSNPs adjuvant and immunogenic peptides in BSA. Accordingly, no signs of hypersensitivity were observed due to the administration of such formulations.
