Abstract

This study aims to investigate the role of Agrobacterium rhizogenes strain EMCC 1744 containing the Ri gene in the induction of hairy roots of Brassicaceae family plants, as well as to evaluate the biological activities of Ri gene-induced secondary metabolites of the Brassicaceae plant extracts, which is achieved by applying an Agrobacterium rhizogenes bacterial suspension on each of five chosen Brassicaceae plant cultures, namely: Brassica campestris, Brassica napus, Eruca sativa, Brassica junceae and Brassica rapa subsp. rapa. This is to allow bacterial Ri gene transfer and integration in the plant genome, and hence, induction of hairy roots. Transformed plants showed higher hairy roots induction efficiency when compared to the wild type. Induction by the Ri gene transfer was confirmed by analyzing the Ri gene expression by means of RT-qPCR technique in all transgenic lines of the 5 Brassicaceae plants. As a result of hairy roots production, secondary metabolites were produced. Total Flavonoid Content (TFC), Total Phenolic Content (TPC), and Total Reducing Power (TRP) percentages in plant extracts were evaluated. The results revealed higher TFC, TPC and TRP levels in all transgenic root extracts, in comparison to that of wild type.
