Abstract

The current work study aimed to Optimize bull sperm quality by using different natural cryoprotectants as anti-oxidants and to evaluate its fertilizing capacity by artificial insemination of female and estimated conception rate.
This current work study was done on Four cattle bulls (aged 3.5-5.0 years) kept at the Animal Reproduction Institute, Central Organization for Veterinary Services, Ministry of Agriculture, Egypt, were selected for semen collection. Then Ejaculates were collected using artificial vagina at weekly intervals for 36 weeks. Ejaculates fulfilling minimum sperm motility (70%) and normal sperm morphology were pooled in order to have sufficient semen for a replicate and to exclude the bull effect. Semen was processed as follow:
1-Experiment 1: Effect of Tris extender enriched with Turmeric extract on cattle semen cryopreservation. 2- Experiment 2: Effect of Royal Jelly Tris-enriched extender on cattle semen cryopreservation. 3- Experiment 3: Effect of Tris extender enriched with Wheat germ extract on cattle semen cryopreservation.
The assessment was implemented post cooling and on freeze- thawed bull spermatozoa. Frozen straws were thawed at 37°C/ 1 minute. The parameters studied were subjective semen characteristics percentage of (motility, alive, abnormality, hypoosmotic swelling test (HOST) and acrosome status).
Results Showed that Tris extender enriched with Turmeric enhanced sperm motility in all concentrations and amelioration in sperm membrane integrity
(HOST) in all concentrations if compared to the control and the best was in TT2 in post-thawing. Conception rate was the best in all concentrations if compared to the control & the best was TT3.
Royal Jelly Tris-enriched enhanced, Sperm motility of post-thawed semen exhibited that, the superior was given with the first and second concentrations of royal jelly while, the lowest was at the last two concentrations. Conception rate was the best in TR1and TR2.
Tris extender enriched with Wheat germ enhanced, Sperm membrane integrity (HOST) in TW1, while acrosome integrity was improved in TW1 and TW2. Motility was also improved in TW1, TW2 and TW3.
Post- thawing findings showed significant improvement in sperm motility in TW1, TW2 TW3 if compared to TW4 and the control.
