Abstract

Infections with Campylobacter species are thought to be spread primarily by poultry. The aim of this study was to find out how common Campylobacter jejuni is in broiler and laying chickens, as well as humans, and to look into the virulence gene profile of C. jejuni.
Two hundred broiler chicken samples (intestine, meat, liver), 160 laying chicken samples (intestine, cloacal swabs and eggs) and 75 human stool samples were collected from some governorates in Egypt. The samples were microbiologically investigated, and the C. jejuni isolates were confirmed biochemically. Some of the isolates were also confirmed by PCR targeting the mapA gene specific to C. jejuni. All isolates were subjected to cadF virulence gene detection using PCR while DNA sequencing of 3 positive samples was performed.
17.33%, 17% and 11.87% of human stool, broiler chicken and laying chicken samples, at the same order, were positive, with a total of 66 Campylobacter jejuni isolates (15.17%). Only ten C. jejuni isolates (15.15%) carried cadF virulence gene (7.69%, 20.58% and 10.52% of human stool samples, broiler and laying chicken respectively). Phylogenetic analysis showed that two of the isolates from chicken meat and intestine have high homology with other C. jejuni isolates from human stool. Moreover, amino acid sequence alignment showed a change in these isolates of zoonotic importance.
C. jejuni isolated from poultry constitutes a public health concern because of the harboured virulence gene.
