Abstract

The purpose of this study was to investigate the impact of using black plum (Prunus salicina) and black cherry (Prunus cerasus) juices in the alleviation of the side effects resulting from multiple sclerosis (MS) in rats. Chemical composition and phytochemical screening of the two fruits were carried out in addition to the estimation of the antioxidant activity of the fruit juices in vitro using DPPH scavenging activity and reducing power. The biological experiment used forty-eight albino rats (weighing 135 to 145 gd. After the adaptation period, the rats were divided into eight groups (6 rats each). One of them was considered a negative control (normal control). The remaining groups were split into two main parts as follows; Part 1(normal rats) consists of 3 normal groups, which were treated daily with one of the three juices (black plum, black cherry, or their mixture) in a dose of 15 ml/kg body weight. Part 2 (cuprizone-induced MS rats) consists of the four groups treated with a standard diet with cuprizone in a dose of 3 g / kg diet. One of the injured four groups was left as a positive control (MS group), while the other three groups were treated daily with the juices of black plum, black cherry, and their mixture (1:1 v/v) in a dose of 15 ml/kg b.wt., using a stomach tube. The weight gain of the rats and feed efficiency ratio was calculated. At the end of the experiment, the rats were sacrificed, and their blood and some organs were drawn for analysis. The results showed that the weight gain and feed efficiency ratio decreased significantly in the positive control as compared to the normal control. At the same time, significant increases in their values were seen in the three treated MS groups compared to the positive group. Regarding liver and kidney functions, the findings showed significant decreases in both ALT and AST and a substantial increase in serum albumin in the MS groups treated with the juices. Also, a significant reduction in their serum creatinine was observed compared to the positive control. As for lipid profile, significant decreases in serum levels of total cholesterol, triglycerides, and LDL-c and a significant increase in HDL-c were noticed in the MS groups treated with the juices. The results revealed that all the tested inflammation parameters decreased significantly. At the same time, the blood immunoglobulins (IgG and IgM) increased in all the MS-treated groups compared to the positive control group. Regarding the antioxidant activity, a significant decrease in malondialdehyde (MDA) and a significant increase in total antioxidant capacity (TAC) have been achieved by the used juices in the MS groups. The histopathological examinations showed significant improvement in the brain tissues and sciatic and optic nerves. It can be recommended to use black plum and black cherry fruits as a perfect nutritional source for those suffering from neurological diseases or inflammations.
