Abstract

The present study designed to evaluate the efficacy of NCur in ameliorate the adverse effects of Al2O3 and Al2O3-NPs on blood, bone marrow, liver and brain of male mice. Seventy two healthy male albino mice (CD1) weighing 22-26 gram that were bought from Theodor Belharz Institute, Cairo, Egypt. They were kept in cages in the Animal House, Zoology Department, Faculty of Science, Assiut University at room temperature (25±2°C) with normal 12 h light/12 h dark cycle for 2 weeks to be acclimatized before starting of the experiment. All animals were fed with standard chow ad labitium, and drunk from tap water. Mice were divided into five groups and doses were orally administered daily for four weeks. group 1 served as control administered with solvent, groups 2 and 3 administered with Al2O3 and Al2O3-NPs, respectively. group 4 administered with Al2O3 and NCur and group 5 administered with Al2O3-NPs and NCur.
After the end of experiment blood samples taken from the jagular vein during mice scarification, bone marrow aspiration smears were commonly isolated from the femur; Smears were immediately prepared for cytological and dysplastic evaluation within 2–3 minutes of the animal’s death, specimens from the liver and brain were quickly taken and sliced into small pieces. Small pieces fixed in formal alcohol for histological study and the other pieces were stored at -80 0C for biochemical and molecular studies.
Complete blood count, electrophoresis for serum protein, types of bone marrow cells, some parameters of oxidative stress and AChE activity, the expression of P53, NRF2 and HSP70 by western blot analysis and pathological observation for brain and liver were investigated.
