Abstract

The present investigation was performed to evaluate the impact of antifungal application on the secretion of lipases and proteases by Candida catenulata. To optimize, extract, purify and characterize the virulent secreted lipases by C. catenulata. To perform in vitro antifungal treatments to C. catenulata and to study their cytological impact on the fungal cells.The molecular identification using 26S ribosomal RNA indicated that the Candida species used in this study was Candida catenulata strain QD16.1. Treatment of C. catenulata with different antifungals revealed that fluconazole (F) was the most inducer to the virulence factor lipase followed by itraconazole (I), then miconazole (M), while combination of (F) and (I) synergistically inhibited lipase induction, so it reduces Candida virulence. Considering proteases, the treatment of C. catenulata with the same antifungals revealed that miconazole (M) was the most inducer to it. The result of the present study showed that Candida catenulata produce much higher lipase activity in presence of fluconazole than protease enzyme. So, optimization of fluconazole treated C. catenulata for lipase production was carried out. Maximum lipase production was obtained at 37{u00B0}C after 3 days incubation at pH 5.5. Dark agitation stimulated growth and lipase production. Medium composed of tween 80 (ml/l) as carbon source, yeast extract (g/l) as nitrogen source, CuSO4.5H2O proved to be optimum for maximum lipase
