Abstract

L-Asparaginase is a crucial enzyme in the treatment of acute lymphoblastic leukemia (ALL) and in acrylamide reduction in heat-treated food products. Thus, the exploration of microbial Lasparaginase free of glutaminase and urease activity as to overcome their adverse safety concerns is of great interest in both medical and food applications. In this study, a novel gene of L-asparaginase free of glutaminase activity (CobAsnase) from halotolerant Cobetia amphilecti AMI6 was cloned and over-expressed in Escherichia coli and characterized. Pre-treatment of potato slices by CobAsnase prior to frying results in 81% reduction of acrylamide compared to untreated control. Also, L-asparaginase free of glutaminase and urease activity was reported for the first time from two newly isolated fungal strains, Aspergillus welwitschiae DH_MS11 and Fusarium arcuatisporum DH_LsRz3. Optimization of the cultural conditions resulted in 2-folds increase in Lasparaginase yield produced by A. welwitschiae DH_MS11. Finally, the investigated Lasparaginases could be potential candidate in cancer treatment and food applications
