Abstract

Background and aim: Rheumatoid arthritis (RA) is the most prevalent autoinflammatory joint disease that results in a considerable burden on both patients and society. The management of rheumatoid arthritis and its complications is a humanitarian and socioeconomic priority. Objectives: To investigate the potential role of cannabinoid receptor 2-agonist (JWH- 133), in rats injected with complete Freund{u2019}s adjuvant in model of adjuvant induced rheumatoid arthritis. Materials and methods: Male albino strain rats weighing (180-200g) were used. The obtained data of the present study used dexamethasone orally daily for 1week (1.5mg/kg), the other drug used was cannabinoid receptor 2 agonist (JWH133) in a dose of 4mg/kg I.P. injection daily for 1week. Rheumatoid arthritis was induced by CFA in a dose of 0.1ml single subplanter injection of right hind paw. After induction of rheumatoid arthritis dexamethasone, CB2 agonist (JWH133) and combination regimen were administrated on day 15 for 1 week. Paw volume of right hind paw was measured just before induction of rheumatoid arthritis, on the day 15(the peak of paw edema and before ttt) and on day 21(after ttt). At the end of the experiment, the following parameters were measured in the serum: RF, MDA and IL-10. Histopathological examination of knee and ankle joints was performed in all groups. Histopathological score were estimated also in all studied groups. Results: normal rats that received saline and DMSO showed insignificant changes of all measured parameters. Rheumatoid arthritis CFA untreated group3 and group 4 showed significant increased in paw edema, elevation of rheumatoid factor, MDA and histopathological score and reduction in IL-10,cartilage thickness and trabecular thickness
