Abstract Rheumatoid arthritis (RA) is an autoimmune disease, where the normal joint tissues attacked by body’s immune system, causing their inflammation. Detecting the expression of CD antigens is supposed to be novel diagnosis methods. Therefore, the aim of current study was to investigate the importance of 2 markers (CD69 and CD73) on peripheral blood lymphocyte cells in the diagnosis of RA; using flowcytometry technique. Patients and methods: This study was carried out on 76 subjects divided into 51 patients with RA and 25 healthy subjects (control). The patients group includes 51 adult patients suffering from rheumatoid arthritis (RA). They were selected from Rheumatology Clinic of Kasr Al-Ainy Hospital, Cairo University in the period from April to September 2017 (a written consent was taken). All subjects were diagnosed clinically according to the 2010 American College of Rheumatology (ACR). The patients were classified according to the presence or absence of rheumatoid factor (RF) into two groups, Seropositive RF group and Seronegative RF group, then the patients were reclassified according to the presence or absence of Anti-cyclic citrullinated peptide (Anti-CCP) into two groups, Seropositive Anti-CCP group and Seronegative Anti-CCP group. The expression of CD69 and CD73 were quantified using flowcytometric technique. Results: In the present study, the expression of CD69 on lymphocyte cells of whole RA patients group were significantly higher than that of healthy control subjects group, while the expression of CD73 on lymphocyte cells of whole RA patients group were significantly lower than that of healthy control subjects group. Conclusion: The expression of CD69 on Lymphocyte cells were significantly elevated in the R.A patients group than that in healthy control group. A reverse pattern was obtained with the expression of CD73 that was significantly decreased in the RA patients group than that in the healthy control subjects group.
