Abstract Background: Genetic factors play an important role in determining the susceptibility to ischemic stroke, two variants in pre miRNA sequences rs2910164 and rs3746444 within miR-146a and miR-499 respectively are studied for their association with ischemic stroke while this relationship are still conflicting and inconclusive. Aim of the work: The aim of this study was to investigate the possible association of miR-146a and miR-499 genetic sequence variation with the risk of ischemic stroke and to assess the possibility of using these variants in risk stratification of atherosclerotic ischemic stroke. Subjects and Methods: A case{u2013}control study was conducted to evaluate the frequencies of both miR-146a C> G polymorphism and miR-499 A>G polymorphism allele variants using real-time PCR TaqMan probes. One hundred ischemic stroke patients recruited from stroke unit of Kasr Al Ainy Hospital and 100 age and sex-matched control group with more than one risk factor for ischemic stroke with no evidence of cerebrovascular disease, ischemic heart diseases or peripheral arterial diseases were included in the current study. Results: the GG genotype and G allele frequencies of rs3746444 within miR-499 A>G were associated with increased risk of ischemic stroke while, GG genotype of rs2910164 within miR-146a C> G was not associated with risk of ischemic stroke in studied population |
