Abstract

This work is concerned with synthesis and optimization of molecularly imprinted polymer specific for nadifloxacin. First, twelve molecularly imprinted polymers were synthesized and optimized using nadifloxacin as a template with different ratios of methacrylic acid and/or 4-vinyl pyridine as a functional monomers and ethylene glycol dimethacrylate as a cross-linker. The synthesized polymers that showed the best binding behavior were MIP₁(1:4:20) (template: methacrylic acid: ethylene glycol dimethacrylate) and MIP₂(1:2:2:20) (template: methacrylic acid: 4-vinyl pyridine: ethylene glycol dimethacrylate). Using further investigations, MIP₁showed higher binding capacity and selectivity than MIP₂. The solid phase extractionprocedures were optimized using MIP1 as a sorbent.The studied factors were: polymer weight, cartridge capacity, loading, washing and elution solvents, cartridge selectivity and cartridge reusability. The optimum procedures were using: 80 mg polymer, methanol as a loading solvent, water: acetonitrile (80:20; v/v) as a washing solvent and methanol: acetic acid (90:10; v/v) as elution solvent. Then, the optimized cartridge was used for specific extraction of nadifloxacin from real human plasma samples
