Abstract

This study proposes nanoemulsions (NE)-based Trojan particles made by entrapping 30.3± 0.29 nm NE in carbohydrate-based microparticles via spray drying. This work focused on optimization of maintenance of the nanocarrier colloidal properties, solid state properties and suitability of Trojan particles for pulmonary deposition. Experimental design was used to study the effect of inlet temperature, ethanol concentration in feed solution and feed solution rate on particle size and product yield of the resultant dry powders. The presence of ethanol had a significant impact on the size, morphology, and density of microparticles and their solid-state properties. Spray drying did not destroy the NE and the size after reconstitution was 117.53 nm. The lactose monohydrate/L-leucine based Trojan particles were readily dispersed as aerosols with mass median aerodynamic diameters between 5.25 and 7.35 μm using a dry powder inhaler. Our research includes an in vivo study to demonstrate the effect of this newly developed formulation on the treatment of allergic asthma (OVA albumin induced asthma) when compared to the market dosage form, as well as the effects of the excipients used in the nanoemulsions and microparticles on lung surfactants. In conclusion NE-trojan particles were successfully prepared and were suitable for pulmonary drug delivery.
