Abstract

BC is a devastating problem leading to morbidity and mortality in developed countries.In Egypt reports showed that BC was detected in (29.7%) among women cancer and represent (22.2%) of death. Breast cancer can be completely cured if it is detected early, but most cases are discovered in the later stages.The present study was planned to evaluate the effect PD-L1, CD20, and CD8 as early predictor biomarkers for breast cancer in Egypt. Blood and tissue samples were used from 50 breast cancer patients, without receiving any treatment or surgical intervention, and this was done with their approval and under the supervision of : Prof. Sameh Roshdy Abdel Aziz, Professor of Oncology Surgery at Mansoura University Oncology Center. In addition to, 15 blood samples from healthy people, as a control group for comparison .The results showed : A significant increase in PD-L1 in patients’ blood samples compared with control. Extreme significant difference in CD20 level in patients’ blood samples compared with control. A dramatic increase of PD-L1 level and a slight increase of CD20 in ILC than IDC in tissue samples. A slight increase in PD-L1 and CD8 in IDC than ILC in blood samples.Immunohistochemical results reflected a corresponding increase of PD-L1 level with the different stages of BC, where the expression increase in late stages within neoplastic glandular cells. Western Blot results, compared with control, blood and tissue samples expressed 1.6 folds and 1.2 folds of CD20 protein signal.CD8 expressions in blood and tissue samples were 1.4 and 1.1 times than that of control respectively.PD-L1 expression level was recorded 1.3 folds for blood samples and 1 fold for tissue samples compared to control.q-PCR gene expression show that all patients reflected varied PD-L1 gene expression when compared to control.This indicates the effectiveness of using PD-L1 gene expression as early predictor procedure for BC diagnosis.
