Abstract

Olanzapine is a principal drug used to treat some mood conditions such as schizophrenia and bipolar disorders. It can be used in combination with some other medications to manage depression. This medication can help to relieve hallucinations Olanzapine belongs to a class of drugs called atypical antipsychotics. It works by helping to restore the balance of certain natural substances in the brain.
Olanzapine has a higher affinity for 5-HT2A serotonin receptors than D2 dopamine receptors, which is a common property of most atypical
antipsychotics. Olanzapine is a potent antagonist of the muscarinic M3 receptor, which may underlie its diabetogenic side effects.
Additionally, it also exhibits a relatively low affinity for serotonin 5HT1, GABAA, beta-adrenergic receptors, and benzodiazepine binding sites. The most common side effects of olanzapine are insulin resistance, diabetes and obesity.
Resistin was found to be produced and released from adipose tissue to serve endocrine functions likely involved in insulin resistance. studies demonstrating that serum resistin levels increase with obesity in several model systems (humans, rats, and mice). further research has linked resistin to other physiological systems such as inflammation and energy homeostasis.
The current study was interventional study conducted at the animal house of El-Nahda university. This study was done on 20 male albino rats and they were divided into two groups:
1- Control group: containing 10 rats.
2- Olanzapine group: study group was given olanzapine injections for 28 days.
The result of the present work showed that the mean value of BMI, serum blood glucose (fasting and postprandial), serum insulin level, Serum Lipid profiles (cholesterol, TG, LDL, VLDL) and serum resistin level are higher in olanzapine than control group but serum HDL level was insignificantly lower in olanzapine group.
from these results we can conclude that:
- Olanzapine administration produced some metabolic effects as insulin resistance, obesity and hyperlipidemia.
- Significant higher levels of serum glucose, lipid profile, serum insulin and HOMA-IR index in olanzapine group as compared with control group.
- Significant higher level of serum resistin in olanzapine group than control group.
- Also the result showed a positive correlation between high levels of resistin, insulin resistance and hyperlipidemia induced by olanzapine administration.
