Abstract Development of hybrid drug candidates is well known strategy for designing antitumor agents. Also, cancer chemotherapy resistance is the innate and/or acquired ability of cancer cells to evade the effects of chemotherapeutics and is one of the most pressing major dilemmas in cancer therapy. This work provides a literature survey for nitric oxide (NO); physical, chemical, biological properties as well as biosynthetic pathways. It also includes classification of NO donors, biochemical pathways of NO generation and NO releasing hybrids against hepatocellular carcinoma (HCC). The thesis describes the preparation of 173 compounds (53 reported and 120 new; 72 intermediates and 48 finals) derived from pyrazolo[3,4-d]pyrimidines and commercially available estrone linked with nitric oxide releasing moieties in an attempt for treatment of cancer especially hepatocellular carcinoma. This work is classified as two main parts: a) NO based pyrazolo[3,4-d]pyrimidines and b) NO based estrone nucleus as Cucurbitacin analogs |
