Abstract

A combination of gastroretentive and pulsatile principles offers an advantage that the system can achieve long residence time in the stomach, sufficient for delivering an adequate amount of drug at the right time, particularly in diseases requiring medication during sleeping and awakening. Developed press coated tablet was found to be suitable for gastroretentive pulsatile delivery of freely soluble drugs after reasonable lag time but not suitable for sparingly soluble drugs and for lesser extent for the practically insoluble drugs. The best achieved formula was found to be containing propranolol hydrochloride (freely soluble drug), ludipress® as core tablet filler, coat polymer blend (methocel® K4M / ethylcellulose 100) with polymer to polymer ratio (60 / 40) and core to coat ratio (1:2). Developed core in cup tablet was found to be suitable for gastroretentive pulsatile delivery of sparingly soluble drugs after reasonable lag time but not suitable for freely soluble drugs and for lesser extent for the practically insoluble drugs
