Abstract

The study aimed to investigate the effect of canagliflozin (CAN) or metformin (MET) treatment for 2 and 4 weeks in obese diabetic rat model (D). The results indicated hyperlipidemia, glucose intolerance and disturbed liver and kidney functions as well as alterations in cortex acetylcholinesterase (AChE) activity and monoamines and amino acids neurotransmitters levels in the D group. Treatment by either CAN or MET resisted the metabolism disturbance by recording significant hypoglycemic and hypolipidemic activities, body weight loss and significant reduction in the diabetic alleviated serum values for liver and kidney functions. In CAN treated group AChE significantly decreased compared to that in D and D+MET groups after the two intervals and both treatments showed effects on neurotransmitters levels. Diabetic group revealed severe focal hemorrhage and blood vessels congestion with neuronal degeneration and gliosis in cerebrum. Treatment with CAN revealed severe congestion in cerebral blood vessels and focal gliosis and with MET showed focal gliosis in cerebrum after the two intervals. The effect of both drugs related to their hypoglycemic and hyperlipidemic activities and the effect on cortex may be in canagliflozin related to its receptor structural activity and related to cerebral metabolism in metformin
