Abstract

Human coronaviruses are ancient viruses that were reported to have existed millions of years ago. Surprisingly, an outbreak of a number of human coronaviruses (HCoVs) was reported in the last decade. Some of which caused the mortality of thousands of people worldwide. The problem of finding a potent drug against HCoV strains lies in the ability of finding a drug that stops the viral replication through inhibiting its important proteins. Despite ribavirin was extensively used as a first choice against viruses it has limited efficacy and potential side effects. Therefore, scientists reverted towards the investigation of different drugs, among which are the direct Acting Antiviral (DAA) drugs. This study aims at comparing the binding to HCoV polymerases of four DAA nucleotide inhibitor drugs that are either approved or in clinical trials for hepatits C Virus (HCV). Quantitative structure-activity relationship (QSAR) and molecular docking are both used to compare the performance of the selected nucleotide inhibitors to the performance of each of their parent nucleotides and ribavirin. Results show that the DAA IDX-184 drug is superior compared to ribavirin; a result that was also reported for HCV
