Abstract

The present study aimed to: Assess leptin expression (mRNA) and secretion (protein) in breast cancer patients and the possible proliferation pathway(s) responsible for breast cancer progression. Investigate leptin concentration in plasma of the breast tumor microenvironment and peripheral blood in breast cancer patients. Test correlation between leptin expression and different subtypes of breast cancer (estrogen receptor positive and negative). A total of 44 women diagnosed with breast cancer and 24 healthy donors were enrolled in the present study from Ain Shams university hospitals. We assessed leptin expression from breast cancer tissues on gene and protein levels using real time polymerase chain reaction (qPCR) and immunohistochemistry respectively. Our results showed that leptin was significantly higher in tissue of obese breast cancer samples than in overweight and control samples (p<0.001). ELISA results showed a significant increase (p<0.001) in leptin in obese women. To investigate whether there is any difference in leptin expression between peripheral and breast tumor microenvironment blood of breast cancer patients, we determined the concentration of leptin in plasma from both using ELISA assay. Our results showed a statistically higher levels of leptin in plasma from breast tumor microenvironment blood of obese estrogen receptor positive (ER+) breast cancer patients than peripheral plasma
