Abstract

The present work aimed to estimate the characterization, lethality percentile doses, including the median lethal dose (LD50) of nickel nano-powder (Ni-NP) to the male albino rats, and its accumulation in the brain, liver, kidneys and intestine, following intra-peritoneal injection of ultra-sonicated Ni-NP. In addition, the work aimed to evaluate the role of Ni-NP as oxidative stressor, and its effect on some biochemical parameters. The estimated median lethal dose (LD50) of ultra-sonicated nano-nickel after 24, 48, 72 and 96 h were 3.38, 1.72, 0.87 and 0.39 g/kg b. wt., respectively. The rats-injected with a repeated sub-lethal dose of 0.02g of Ni-NP/kg b. wt. /2 days (=1/20 LD50 at 96 h) over a period of twenty-eight days. The accumulated Ni-NP in the brain, liver, kidneys and intestine were influenced by experimental time (1, 3, 7, 14 & 28 days), injected doses (0 & 0.02 g), types of organs and their interactions. Nickel accumulated in tissues of rats injected Ni-NP showed a positive correlation with the experimental periods and their average were significantly higher than the corresponding controls. In the brain, hepatic and renal levels of malondialdehyde (MDA) and oxidized glutathione (GSSG) showed a positive direct relationship with Ni-NP accumulated in these tissues, whereas glutathione (GSH) and the ratio of GSH/GSSG showed a negative correlation
