Carvedilol (CVD) is non selective Ý-blocking agent. It is a potent antihypertensive agent with a dual mechanism of action. At relatively low concentrations, it acts a competitive beta-adrenoceptor antagonist and a vasodilator, whereas at higher concentrations it is used as a calcium channel antagonist. CVD suffers from low absolute bioavailability (25%) following oral administration; probably due to the limited solubility of CVD in water as well as the considerable first-pass metabolism in the liver. In the current work, buccal formulations of carvedilol were developed in a trial to overcome the first-pass metabolism of the drug, to improve its oral bioavailability and to control the rate of drug release via the buccal mucosa.
