Carvedilol (CVD) is non selective Ý-blocking agent. It is a potent antihypertensive agent with a dual mechanism of action. At relatively low concentrations, it acts a competitive beta-adrenoceptor antagonist and a vasodilator, whereas at higher concentrations it is used as a calcium channel antagonist. CVD suffers from low absolute bioavailability (25%) following oral administration; probably due to the limited solubility of CVD in water as well as the considerable first-pass metabolism in the liver. In the current work, buccal formulations of carvedilol were developed in a trial to overcome the first-pass metabolism of the drug, to improve its oral bioavailability and to control the rate of drug release via the buccal mucosa. The work in this thesis is thus divided into three chapters: Chapter Ї: Formulation and In Vitro Evaluation of Carvedilol Buccal Tablets Chapter ЇЇ: Formulation and In Vitro Evaluation of Laminated Buccal Sponges Loaded With Carvedilol Microemulsions Chapter III: Pharmacokinetic Assessment of Carvedilol Following Buccal Administration of the Best Achieved Formula in Healthy Volunteers
