Abstract

P. aeruginosa is a widespread species in the environment. It has the ability to behave as an opportunistic pathogen and target immunocompromised people, suffering from AIDS, burns, cystic fibrosis or cancer. In this study, biochemical and molecular characterization of bacterial isolate screened from composted pile wastes indicated that it is P. aeruginosa strain TMN01. Optimization of cultural conditions for maximizing the production of exotoxin A (ETA) using statistical design Taguchi system indicated that agitation rate, temperature and interaction between incubation period and beef extract were the most significant variables in ETA excretion by P. aeruginosa. The yielded protein was cytotoxic to Swiss Albino mice in the cytotoxicity assays. Complete purification protocol was used by (NH₄)₂SO₄ precipitation anion exchange by DEAE cellulose and gel filtration by Sephadex G200 and resulted finally in production of purified single band of ETA protein (9 æg ml⁻¹) with 36.8 % cytotoxicity relative to crude ET
