Abstract

Damage to the reproductive system is one of the most devastating effects of chemotherapeutic treatment, which is frequently associated with premature ovarian failure (POF). Therefore, this study was planned to evaluate the therapeutic effectiveness of Adipose-Derived Stem Cells (ADSCs) in a rat model of chemotherapy-induced ovarian failure. Ovarian failure was induced by combined cyclophosphamide/busulfan therapy. Intravenous injection of PKH26 labelled ADSCs was done after 1week of chemotherapeutic treatment. Serum FSH, LH and Estradiol were measured. Animals were sacrificed 1 week and 4 weeks after ADSCs injection. Ovarian specimens were processed and stained with Hematoxylin and Eosin, Masson{u2019}s Trichrome and immunohistochemical staining for PCNA and Bcl-2. Adipose-Derived Stem Cells-treated group exhibited improved hormonal profile, improved ovarian histological architecture and increased folliculogenesis. Collectively, these results suggest that ADSCs are of considerable therapeutic promise for restoring fertility after chemotherapeutic treatment
