Abstract

The objective of the current study is monitoring the circulation of five equine herpesviruses (EHV-1, EHV-2 EHV-3, EHV-4 and EHV-5) among different horse populations. Different field samples were collected from different farms and clinics located in Egypt and screening of the collected samples was done using Quantitative PCR and conventional PCR assays for detection of EHVs followed by nucleotide sequence analysis of the amplified PCR products.A total of 187 samples (including 102 nasal swabs, 40 uterine swabs, 7 coital swabs, 10 tissues and 28 whole blood samples) were collected for molecular screening of EHVs by different PCR assays and 20 serum samples for detection of EHV specific antibodies by ELISA. Molecularly identified samples were selected and isolated on MDBK cell culture. In parallel isolation of EHVs was carried out on CAM of 11 days old SPF-ECEs.Screening of extracted DNA of prepared field samples by real time PCR and Pan herpesvirus PCR revealed that 114/187 (55%) samples were positive for EHVs. Typing of positive samples by PCR assays using specific primers and probes showed the percent of each type of EHVs as follow: EHV-1 (71/187; 38%), EHV-2 (65/187; 31%) , EHV-3 (3/187; 1%), EHV-4 (15/187; 8% ) and EHV-5 (38/187; 20% ).The molecularly identified field samples were selected and successfully propagated on MDBK cell lines and displayed positive cytopathological changes as aggregation of cells, focal rounding and finally, cells detached. In parallel successive isolation of the virus in field samples was conducted on CAM of SPF-ECEs producing pock lesions on CAM.Phylogenetic analysis of the obtained EHVs sequences in the current research showed that they were similar to foreign strains.
